Après avoir déménagé à Baltimore dans le Maryland, Lawrence Gilliard Jr. rejoint la Juilliard School pour étudier la musique classique, aux côtés de Jada Smith et Tupac Shakur, et finit son cursus en comédie. A 20 ans, le jeune homme obtient son premier rôle dans Straight Out of Brooklyn dans lequel il joue un jeune défavorisé prêt à tout pour s’en sortir. Souvent relégué à des rôles de voyous, l’acteur enchaîne les apparitions dans des séries policières telles que New York Section Criminelle, Homicide, New York Undercover ou Les Experts : Manhattan, mais c’est avec Sur Ecoute qu’il se fait connaître du grand public. D’Angelo Barksdale, immoral et cruel, contrôle le marché de la drogue dans le quartier Ouest de Baltimore. Côté ciné, ses personnages notables incluent un adolescent du ghetto amoureux dans Lotto Land, un joueur de football honnête dans The Waterboy et un membre des Deads Rabbits dans Gangs of New York de Martin Scorsese. Il reprend le chemin de la télévision dans Numb3rs, Lie To Me, American Wives, Friday Night Lights ou Longmire jusqu’à The Walking Dead. En effet, en 2013, il prête ses traits à Bob Spookey, alcoolique désœuvré et petit ami de Socha, qui reprend goût à la vie en rejoignant Rick et sa bande dans la saison 4..
